چکیده

A method is given for selecting epithelial cells directly from primary mammary tumors in Methocel suspension culture. The frequency of colony-forming units in primary tumors was ap proximately 10~4. Colonies grew by cell division; formation and growth of colonies was cell density dependent. Five Methocel isolates were established in monolayer culture and character ized. Two were epithelial, evidenced by functional occluding junctions. The other three were not typed in vitro, although they formed carcinomas in vivo. All were subtetraploid by passage 10. There were variations in ability of the five Methocel isolates to redone in suspension that appeared to be due to the evolution of anchorage-dependent variants during their growth in monolayer culture. These variants could be purified by limiting dilution plating on solid substrates. The five Methocel isolates and their derivative variants were used to determine correlations between transformation markers and tumorigenicity. Only three Methocel-derived sublines of nine tested, including two recloned in Methocel, were tumorigenic at all when inoculated in two sites of three syngeneic hosts, one athymic. The other six were nontumorigenic. The tumorigenic sublines were less tumorigenic than uncul tured cells of parent tumors or parent tumor cells grown in primary monolayer culture. Thus, anchorage-independent growth is not a reliable marker for the tumorigenic mammary phenotype. No correlation was found between two other "con tact-related" transformation markers, rapid growth rate and monolayer overgrowth, and tumorigenicity. The three transfor mation markers were expressed independently. Both tumori genic and nontumorigenic sublines expressed mammary tumor virus antigens M.W. 28,000 protein and M.W. 52,000 glycoprotein, although only a minor fraction of cells contained the M.W. 52,000 glycoprotein. These data emphasize the hetero geneity of phenotypes in mammary tumors as well as differ ences between fibroblasts and mammary epithelium in models of neoplastic transformation.

Detection of Mouse Cytomegalovirus in Adenocarcinoma Bearing Razi/A Mice: Molecular and Pathological Studies

Despite a lot of research, the etiology and progression of breast cancer remain incompletely understood. Recently, human cytomegalovirus (HCMV) was reported as a risk factor for breast cancer. The aim of this study was to know whether breast cancer could be caused by cytomegalovirus or not? In this experiment seventeen samples of RAZI/A mice with spontaneous breast cancer were being gathered fr...

بررسی توان القای مرگ سلولی سلول‌های میو اپی تلیال موشی بر رده‌ی سلولی کارسینومای پستان

Background and Objective: Breast tumors include the heterogeneous population of various cells such as luminal and myoepithelial cells which can play a significant role in the development and/or the inhibition of breast cancer. It has been observed that paracrine and autocrine activities of myoepithelial cells can limit the mammary tumors via angiogenesis inhibition and apoptosis induction. Ther...

Interaction of primary mammary bovine epithelial cells with biofilm-forming staphylococci associated with subclinical bovine mastitis

Background: Staphylococci are recognized worldwide as one of the most important etiological agents of bovine mastitis due to their virulence factors such as their ability to penetrate inside mammary epithelial cells and their ability to form biofilm. Aims: The objectives of this study were to establish a model of primary mammary epithelial cells origin...

Stroma by, Malignant Human Breast Cancer Epithelium Spontaneous Fusion with, and Transformation of Mouse

Adenocarcinoma cells from the pleural effusion of a patient with breast cancer were injected into the mammary glands of nude mice and grown into solid tumors. A cell line derived from these tumors expressed A-smooth muscle actin but not human cytokeratin 7, indicating ‘‘activated’’ stroma of mouse origin. Cells in mitosis exhibited mainly polyploid mouse karyotypes, but 30% had mixed mouse and ...
